The opportunity and challenge for the 21st century life science research
The life science research in 21st century is an opportunity as well as a challenge for each science researcher.

The research and application of protein and peptides is a major direction in the biotechnology field. Separation and purification are both involved in protein and peptide research despite to the lack of structure or the function information.

Recently medicinal biotechnology has been the core motivation during the development in biotechnology field. Due to the limits of technology development investment and relative weak research and development ability of enterprises, the separation and purification in biotechnology pharmaceutics has hindered its development.

The chromatographic separation materials, separation technology, efficient separation and purification solutions for protein and peptides have been presented by Acchrom.

Bio-C18 Reverse-phase Chromatography Column
RPLC has been widely applied in peptides separation for a long time owing to its good selectivity, wide applicability, high repeatability and easy selection of mobile phase. Although most low molecular weight proteins and peptides can be separated in RPLC, RPLC technology is insufficient for the separation of peptides owing to the varieties of peptide chains, acid-base property, hydrophobicity, complex sources and compositions.

Polar hydrophilic groups have been introduced in reversed-phase bonded silica based on the unique "polar-copolymerized approach" by Acchrom. It produces steady surface properties, hydrophobic and polar selectivity in RPLC. It offers high selectivity and efficient separation in RPLC and prolonged the life of columns effectively.

Bio-C18 column presents sharp chromatographic peaks and high resolution for LC/MS analysis.

Series of HILIC Mode Columns

HILIC, as a supplement to RPLC, has been generally applied in peptides separation in recent years. HILIC has good orthogonality with RPLC due to their distinct separation mechanism. Acchrom Technologies Corp. has developed a series of silica gel-based materials with the functions of zwitterion (Bio-Amphion) and amide (Bio-Amide). These materials can be applied in the separation and purification with high resolution of peptides, which can provide technology basis for the quality control and purification of peptide pharmaceuticals.

The separation mechanisms of HILIC and RPLC are completely different and they may have superior orthogonality, which is favorable for building two-dimensional HPLC system of peptides in order to solve complicated separation and analysis issues of peptide samples. A first dimension fraction on RPLC column is injected again into the second dimension on HILIC column (Bio-Amphion). Many chromatographic peaks can be separated and further leads to the discovery active peptides.

'Bio' series of Mixed-mode columns
Separation of protein and peptides especially for complex ones cannot be realized by single RPLC mode; however, mixed-mode chromatography may provide unique separation owing to its combination of two or more separation mechanisms.

A series of mixed-mode columns that combines both RP and ion-exchange properties have been developed for protein and peptides separation. Better selectivity achieving can be achieved than conventional C18 stationary phase, which is appropriate for various properties of peptides and protein separation.

Better Selectivity and Peak Shapes
Ion-exchange interaction and polar selectivity are introduced on the basis of hydrophobicity selectivity —— better selectivity can be performed on Bio-RP/WCX than conventional C18 column.

Peptides Clustering Effect to Achieve High Performance Enrichment and Separation
The existence of surface charge interaction make Bio-RP/WCX column equip with peptides clustering effect.

Acidic peptides with negative charge and basic peptides with positive charge are eluted from Bio-RP/WCX column based on ion-exclusion and ion-exchange interaction respectively. It leads to clustering separation of peptides.

  

Protein Separation Technology
Generally the lack of the hydrophilicity on stationary phase surface results in strong adsorption of proteins when conventional RPLC column is used to separate proteins. It will lead to changing of retention time, selectivity and low column efficiency.

Polar hydrophilic groups have been introduced in reversed-phase bonded silica based on the unique "polar-copolymerized" proprietary technology by Acchrom. Series of Bio-C8 RPLC columns have been developed by improving the surface hydrophilicity of the separation materials. Better peak shapes and selectivity of proteins can be obtained on Bio-C8 columns and the column life can be prolonged as well.
