Importance of Oligosaccharide
Carbohydrates are probably the most extensive compounds in the biological world, which are considered as structural materials and energy sources for a long time. Recently, carbohydrates especially oligosaccharides have gained more attention due to their biological functions. Oligosaccharides that combine with proteins to form glycoproteins, play important roles in life. Glycosylation is one of the most common post-translational modifications (PTM) of proteins. More than half of the proteins in eukaryotic cells are glycosylated. Meanwhile, glycosylation also occurs in the intracellular regions. Glycosylation not only has vital impact on protein folding, localization, trafficking, solubility and antigenicity, but also play key roles in cell to cell interactions.

Current Situation and Demand for Oligosaccharide Separation
Separation of oligosaccharide is the starting point for a deeper understanding of the structure−function relationship of oligosaccharide. It is also significant for quality control of products containing oligosaccharide.

Hydrophilic interaction liquid chromatography (HILIC) is very suitable for the efficient separation of oligosaccharides owing to its characteristics and superiority. The available HILIC stationary phases for separating small pharmaceuticals include hydroxyl-, zwitterionic- or polymer-modified silica materials. However, due to high polarity, and the structural variety of isomeric structures, the separation technology and materials dedicated for separation of oligosaccharides are limited.

In order to accurately determine the site-specific glycosylation information and the structure of glycan, the enrichment of glycopeptides with high coverage before MS analysis is of great importance.
